Project Description

Research will be conducted in role of miRNAs in the Control of Oocyte Reserve in Fish. This thesis is part of a research project that seeks to uncover the basic workings of the egg supply in fish ovaries, which produce mature eggs during each reproductive cycle. This study aims to help us understand the natural changes in fish that are important for things like protecting wild fish populations and making fish farming better. This project will be co-supervised by Violette Thermes (LPGP, INRAE) and Frédérique Clément (MUSCA, INRIA).

The researcher will employ a variety of techniques in their work. These include 3D imaging through confocal microscopy, which will be conducted using the LPGP 3D imaging platform. They will harness open-source Deep Learning algorithms for image analysis to extract valuable insights from their data. Additionally, histology techniques such as in situ hybridization (utilizing RNAscope technology) and immunohistochemistry, involving antibodies or nanobodies, will be utilized to examine tissue samples. To delve into gene expression patterns, the researcher will turn to transcriptomics approaches, specifically RNA-seq. These techniques collectively form the toolkit for their research, enabling a comprehensive exploration of their objectives.

Hypothesis of the Project

The current hypothesis is that, similar to mammals, the regulation of the egg supply in a female fish’s lifetime involves paracrine mechanisms mediated by factors like the anti-müllerian hormone (AMH). In our laboratory research, we have also identified two specific microRNAs that are crucial for maintaining a female fish’s ability to lay eggs in each reproductive cycle. Based on these findings, we propose that these two microRNAs, in addition to AMH, play key roles in managing the egg reserve and controlling the overall reproductive capacity of female fish.

Objective of Project

  • To understand the role of AMH and two miRNAs in forming and renewing the egg reserve in fish.
  • To conduct in vivo experiments in specially bred Medaka fish lines (including KO and transgenic) and create new fish lines using CRISPR/Cas9 technology.
  • To gain insights into the mechanisms behind this process with implications for preserving wild fish populations and improving fish farming practices.
